The Der p 2 ELISA kit is used for measuring the major allergen Der p 2. The Dermatophagoides pteronyssinus protein Der p 2 is one of the major house dust mite allergens. Using this Enzyme-Linked Immunosorbent Assay (ELISA) kit the concentration of this allergen in solution can be determined. This kit provides sufficient antibodies and Der p 2 standard to perform ELISA experiments on five 96-well plates. Until now there was no specific ELISA for Der p 2 available. For the group 2 allergens there is the group 2 (Der p 2 and Der f 2) ELISA kit available but this is not a complete group for all the house dust mites. With the introduction of this unique Der p 2 kit there is finally a solution for measuring the D. pteronyssinus Der p 2 allergen.

The ELISA kit is sufficient for the coating of 5 plates.

Contents of the Der p 2 ELISA kit

  • 1 vial with capture antibody (white vial with blue endcap)
  • 1 vial with detection antibody (brown vial with brown endcap)
  • 5 vials with Der p 2 standard (white vial with wite endcap)

All components of the kit are supplied in freeze-dried (lyophilized) form in 0.5 ml vials. It has a longer shelf life than the regular ELISA kits which will be delivered in liquid form. One of the advantages of our kit is that the Der p 2 standard will be delivered in 5 separate vials, this way you can use 1 vial with the right amount for each plate. Just dissolve the material in your buffer of choice and you are ready to coat the plate.

Standard Der p 2

The Der p 2 standard for this ELISA kit is based on purifying the natural proteins. For the Der p 2 kit this of course is purified natural Der p 2. We have weighed several batches of natural purified proteins (>98 %) which we have used to determine the standard. To our understanding this is the most elegant way  to determine the amount. By this we have a trusted standard and a very reliable ELISA kit.

In order to quantify Der p 2 in house dust mite extracts a reliable method for Der p 2 concentration determination is required. The Der p 2 ELISA assay is used to measure the concentration of Der p 2 in house dust mite extracts. Finding a defined Der p 2 standard, it has become acceptable to use a complex standard to determine Der p 2 concentrations in house dust mite extracts. However, our natural purified Der p 2 can be used is an accurate standard by weight.

Natural Der p 2 is purified from a Dermatophagoides pteronyssinus extract. We can use the natural purified antigen Der p 2 to an amount we can weigh. The concentration of the purified Der p 2 is determined by OD 280 nm. Analysis performed are protein determination (BCA and Bradford method) and SDS-PAGE (CBB and silver staining).

Purifying Der p 2 using biochemical techniques, we have reached a purity of >98 % and we have weighed amounts of different batches (between 5 and 10 mg per batch). By purifying Der p 2 we have achieved our golden standard. Based on the golden standard we can produce standards in amounts which can be used for one ELISA plate.
